Yes you can. Respect of identity isn't contingent on understanding. You can ask for information to help you understand, but you can't demand understanding before granting respect. If you don't believe someone's identity to be authentic, then don't interact with them rather than normalising invalidation.
Would it be better to say "I'd be more comfortable and understanding of your gender identity if you explain it to me in more detail."?
Not really. You can ask someone to explain to help you understand, but they don't owe you comfort or understanding. They should have your support even if you don't understand.
